Human cytomegalovirus (HCMV) antigens pp65, IE1, and glycoprotein B (gB) are the primary immunodominant targets used in the development of vaccines and adoptive immunotherapies to combat CMV infection. pp65 (UL83) is a major tegument protein that acts as the principal target for the host's cellular immune response, specifically CD8+ T cells, while IE1 (UL123) is an essential immediate-early regulatory protein that is also a key target for early T-cell recognition (UniProt P06725, P13202). Glycoprotein B (UL55) is a critical envelope component required for viral fusion and entry, making it the primary target for neutralizing antibodies (UniProt P06473). In clinical settings, these antigens are often combined in multi-antigen platforms, such as the Triplex vaccine, to provide comprehensive protection by stimulating both humoral and cellular arms of the immune system (PMID: 29626057). This approach is particularly vital for immunocompromised patients, such as transplant recipients, where restoring CMV-specific immunity is essential to prevent life-threatening organ disease and viremia.
