Overview

The human double minute 2 protein, commonly referred to as MDM2 or E3 ubiquitin-protein ligase Mdm2, is a nuclear-localized protein that functions primarily as an E3 ubiquitin ligase and is a critical negative regulator of the p53 tumor suppressor pathway. Overexpression or amplification of MDM2 leads to excessive degradation/inhibition of p53, promoting tumorigenesis. Due to its central role in regulating the key tumor suppressor protein p53—and frequent dysregulation in cancer—MDM2 is a major target for anticancer drug development.
