Overview

Human epidermal growth factor receptor 2 (HER2)-derived peptide antigens are specific amino acid sequences derived from the HER2/neu protein, which is frequently overexpressed in various malignancies (Saha et al., Frontiers in Oncology, 2021). These peptides, such as E75 (Nelipepimut-S), GP2, and AE37, serve as tumor-associated antigens (TAAs) that are utilized in cancer vaccines to stimulate the host's immune system (Mittendorf et al., Cancer, 2014). When administered, these peptides are processed and presented by major histocompatibility complex (MHC) molecules on the surface of antigen-presenting cells. This presentation triggers the activation and expansion of HER2-specific CD8+ cytotoxic T lymphocytes and CD4+ helper T cells, which then target and eliminate HER2-positive tumor cells (Patel et al., Journal of Clinical Oncology, 2010). This therapeutic strategy is primarily employed in the treatment of breast, gastric, and ovarian cancers to prevent disease recurrence by establishing long-term immunological memory. Unlike monoclonal antibodies that provide passive immunity, HER2-derived peptide antigens aim to induce a durable, active immune response against the cancer (NCI Drug Dictionary).
