Overview

HER3 (ERBB3) is a receptor tyrosine kinase and member of the EGFR/ERBB family. It functions primarily as a co-receptor due to its impaired kinase activity, forming heterodimers with other ERBB receptors like EGFR/HER1 or HER2/ERBB2 upon ligand binding (e.g., neuregulins). This heterodimerization leads to phosphorylation events and activation of downstream signaling cascades, including PI3K/AKT and MAPK/ERK, which are critical for cell growth, differentiation, and survival. Overexpression or dysregulation of HER3 is implicated in various cancers, making it an important therapeutic target.
