Overview

HER4, also known as ErbB4 or ERBB4, is a receptor tyrosine kinase and a member of the epidermal growth factor receptor (EGFR/HER) family. It functions as a single-pass type I transmembrane glycoprotein with intrinsic tyrosine kinase activity. HER4 is activated by ligands like neuregulins and plays essential roles in normal development and function of various organ systems. Mutations or altered expression/activity are implicated in several diseases, including cancer and neuropsychiatric disorders. HER4 undergoes regulated intramembrane proteolysis resulting in release of two fragments: an extracellular fragment and a cytoplasmic fragment that can translocate to the nucleus to influence gene expression directly.
