The human H19 promoter is a regulatory DNA sequence that controls the expression of the H19 lncRNA. It exhibits tumor-selective activity, being largely silent in most adult tissues but reactivated in many tumors. This selectivity makes it valuable for driving targeted gene expression in cancer therapy. Promoter activity is influenced by DNA methylation, histone modifications, and transcription factor binding. It should be noted, however, that despite its higher activity in tumor cells, expression does occur in normal cells, and this has to be carefully considered for therapy delivery.
