Overview

NAALADL1 (human ileal aminopeptidase, also called N-acetylated alpha-linked acidic dipeptidase like 1 or NAALADase L) is a zinc-dependent metallopeptidase with broad aminopeptidase activity and is highly expressed in the human small intestine, primarily in the ileal brush border membrane[1][4][7]. Unlike the closely related glutamate carboxypeptidase II, NAALADL1 lacks carboxypeptidase and dipeptidyl-peptidase IV type activities and does not act on substrates with both Pro in the P3′ position and Asp or Glu in the P2′ position[1][2][7]. Its main physiological role is associated with the final stages of protein and peptide digestion and absorption in the human digestive process[1][3][4]. It is not currently established as a direct therapeutic target, but its inhibition (e.g., by bestatin) is biochemically characterized[1][3].
