The Human Immunodeficiency Virus 1 (HIV-1) envelope glycoprotein (Env) is the essential machinery for viral entry and the primary target for the humoral immune response. It is synthesized as a gp160 precursor that is proteolytically cleaved into the surface subunit gp120 and the transmembrane subunit gp41, which together form a functional trimeric spike on the virion surface [1, 2]. gp120 mediates initial attachment to the host CD4 receptor and subsequent binding to chemokine co-receptors (CCR5 or CXCR4), while gp41 facilitates the fusion of viral and cellular membranes [2, 3]. As the only viral protein exposed on the surface, Env is the target of broadly neutralizing antibodies (bNAbs) and various entry inhibitors like Fostemsavir and Enfuvirtide [4, 5]. Therapeutic intervention at this target aims to prevent the establishment of infection or reduce viral load in chronic cases by blocking the viral entry pore [3, 5]. The high mutation rate of the Env gene leads to significant antigenic diversity, posing a major challenge for vaccine development and long-term efficacy of monoclonal antibody therapies [2, 3].
