Overview

Human immunodeficiency virus 1 (HIV-1) integrase is a viral enzyme essential for the replication of HIV-1, the causative agent of AIDS. It catalyzes the integration of viral DNA into the host cell genome, a critical step that allows for persistent infection and efficient viral replication. This makes HIV-1 integrase a major target for antiretroviral drug development. HIV-1 integrase performs two main catalytic reactions during viral replication: 3'-Processing and Strand Transfer. By integrating its genome into host chromatin, HIV establishes lifelong infection, evades immune detection, and ensures stable expression and propagation within dividing cells. Integrase is targeted by several classes of antiretroviral drugs known as "integrase inhibitors" or "INSTIs" (e.g., raltegravir, elvitegravir, dolutegravir), which block strand transfer activity.
