Human immunodeficiency virus type 1 (HIV-1) reverse transcriptase (RT) is a multifunctional enzyme essential for the replication of HIV-1, the causative agent of AIDS. RT catalyzes the conversion of the viral single-stranded RNA genome into double-stranded DNA, which is then integrated into the host cell genome—a defining feature of retroviral replication. RT’s activities are critical for converting the (+)ssRNA genome packaged within virions into linear dsDNA. More than half of currently approved antiretroviral drugs target HIV-1 RT due to its essential role in viral replication.
