Overview

The HIV-1 Gag polyprotein is a major structural protein essential for the assembly, budding, and release of new virions from infected cells. It is encoded by the gag gene as a precursor protein (p55) and undergoes proteolytic cleavage to generate key structural components of the mature virus. Gag orchestrates virion assembly, membrane binding, genome packaging, budding, and maturation. Cleavage of Gag by the viral protease is critical for viral infectivity. Mutations within Gag can lead to antiretroviral drug resistance.
