Gag is processed to form the virion's matrix, capsid, nucleocapsid, and associated spacer peptides, governing assembly and budding at the host cell membrane[1][2][4]. Pol encodes the viral enzymes—protease (PR), reverse transcriptase (RT), and integrase (IN)—required for maturation, reverse transcription of RNA to DNA, and integration of viral DNA into the host genome[2][4]. Env is synthesized as gp160 and processed into surface glycoprotein gp120 and transmembrane subunit gp41, which mediate viral binding to and fusion with host cells using CD4 and co-receptors[2][4]. All three polyproteins are crucial for the production of infectious HIV-1 particles and represent validated therapeutic targets, with many approved antiretroviral drugs and vaccine efforts directed at their functions and interactions[1][2][4][5].
