Overview

The Human immunodeficiency virus type 1 peptide-major histocompatibility complex (HIV-1 pMHC) is a molecular assembly consisting of a short viral peptide fragment bound within the groove of a host Major Histocompatibility Complex (MHC) molecule, typically HLA Class I. These peptides are derived from the proteolytic degradation of HIV-1 proteins such as Gag, Pol, or Env within an infected cell and are transported to the cell surface to signal the presence of intracellular infection to the immune system. The recognition of this complex by the T cell receptor (TCR) on CD8+ cytotoxic T lymphocytes is the fundamental step in the cellular immune response against HIV-1. In the context of therapeutic development, the HIV-1 pMHC serves as a highly specific target for TCR-engineered T cells (TCR-T) and bispecific molecules like ImmTAVs, which aim to redirect the immune system to eliminate the latent viral reservoir. However, the high mutation rate of HIV-1 often leads to 'immune escape' where the peptide sequence changes, preventing TCR recognition, and the virus further evades detection by actively downregulating MHC expression on the surface of host cells.
