Overview

Human IMP1-associated desert definitive endoderm long non-coding RNA (HIDEN, also known as IGF2BP1 associated definitive endoderm lncRNA or IADEN) is a cytoplasmic long non-coding RNA specifically upregulated during differentiation of human pluripotent stem cells into definitive endoderm. HIDEN does not affect the maintenance of pluripotency but is required for successful transition to definitive endoderm. Mechanistically, HIDEN interacts with the RNA-binding protein IGF2BP1 (IMP1), facilitating the binding of IGF2BP1 to FZD5 mRNA and promoting its stability. Stabilization of FZD5, a WNT signaling receptor, enhances WNT pathway activity, which is necessary for definitive endoderm differentiation. Depletion of HIDEN impairs endoderm differentiation by destabilizing FZD5 mRNA and reducing WNT signaling. HIDEN, as a "desert" lncRNA (a lncRNA from a gene desert region), is notable because such RNAs are less well characterized but may play unique regulatory roles in cell fate decisions[1][2][3].
