IMA910 is a multi-peptide therapeutic vaccine designed for the treatment of colorectal cancer (Fiedler et al., 2016). It consists of 13 synthetic tumor-associated peptides (TUMAPs) that are naturally presented on the surface of primary tumor cells and metastases by Human Leukocyte Antigen (HLA) class I and class II molecules (Immatics Biotechnologies). Specifically, the vaccine includes 10 peptides restricted to HLA-A*02 and 3 peptides restricted to HLA-DR, ensuring the activation of both cytotoxic CD8+ T cells and helper CD4+ T cells (ClinicalTrials.gov NCT00785122). These peptides are derived from proteins overexpressed in colorectal cancer, such as CEA and TGFBI. By administering these peptides alongside an adjuvant, the vaccine aims to stimulate a robust and specific immune response against the tumor, potentially overcoming immune tolerance (Walter et al., 2012). Clinical evaluation has focused on its efficacy in patients with advanced colorectal cancer, often in combination with immunomodulators like GM-CSF or low-dose cyclophosphamide to enhance T-cell activity.
