The Human leukocyte antigen A*11:01 (HLA-A*11:01)–KRAS G12V neoepitope complex is a specialized molecular target for cancer immunotherapy, consisting of a mutated peptide fragment from the KRAS protein (specifically the G12V mutation) presented by the HLA-A*11:01 MHC Class I molecule (PubMed: 31413067). KRAS is a member of the RAS GTPase family that regulates cell growth; mutations at codon 12, such as the substitution of glycine with valine (G12V), lead to constitutive activation and are primary drivers in pancreatic, colorectal, and lung adenocarcinomas (NIH: GTR000500348). This specific neoepitope complex is absent on healthy cells, providing a unique window for high-precision therapeutic intervention. Current drug development efforts focus on T-cell receptor (TCR) engineered T-cell therapies and bispecific antibodies designed to recognize this complex with high affinity and specificity (Affini-T Therapeutics, 2023). By binding to this complex, these therapies trigger a targeted immune response, leading to the selective destruction of tumor cells while minimizing damage to normal tissues.
