Overview

The Human leukocyte antigen A*11:01 presenting Kirsten rat sarcoma virus oncogene homolog (KRAS) mutant peptides complex is a highly specific neoantigen target for cancer immunotherapy (Wang et al., 2016, JCI). HLA-A*11:01 is a common MHC Class I allele, particularly prevalent in East Asian populations, while KRAS mutations like G12D and G12V are key drivers in pancreatic, colorectal, and lung cancers (Prior et al., 2020, Cancer Research). When mutant KRAS proteins are processed, their peptides are presented by HLA-A*11:01 on the tumor cell surface, creating a unique target for T-cell receptors (TCRs). Therapeutic strategies include TCR-engineered T-cell (TCR-T) therapies and cancer vaccines, such as ELI-002, which aim to stimulate a precise immune response against these tumor-specific markers (Bear et al., 2021, Cancer Cell; Elicio Therapeutics, 2024). This approach minimizes damage to healthy tissues, which lack the specific mutation-HLA combination. However, challenges such as HLA downregulation and the risk of cytokine release syndrome remain significant considerations in clinical development.
