Overview

Human leukocyte antigen A*24:02 (HLA-A*24:02) is a Major Histocompatibility Complex (MHC) class I molecule that plays a critical role in the cellular immune response by presenting intracellularly derived peptides to CD8+ cytotoxic T lymphocytes (CTLs). The Cell division cycle associated 1 (CDCA1) protein, also known as NUF2, is a component of the NDC80 kinetochore complex essential for chromosome segregation during mitosis (UniProt: Q9BZD4). CDCA1 is significantly overexpressed in a variety of malignancies, including non-small cell lung cancer, gastric cancer, and colorectal cancer, while its expression in normal tissues is restricted primarily to the testis, making it an ideal tumor-associated antigen (PubMed: 16452213). The specific presentation of the CDCA1-65 epitope (sequence: VYGIRLEHF) by the HLA-A*24:02 allele is a key target for cancer immunotherapy, particularly in East Asian populations where this allele is highly prevalent (PubMed: 21830315). Therapeutic strategies targeting this complex include peptide-based vaccines, such as OTS111, and the development of T-cell receptor (TCR) engineered T-cell therapies designed to recognize and eliminate tumor cells presenting this specific antigen (ClinicalTrials.gov: NCT01223235). These interventions aim to stimulate or provide the immune system with the necessary tools to identify and destroy cancer cells while sparing healthy tissue.
