The Human leukocyte antigen A*24:02 (HLA-A*24:02) presenting NY-ESO-1 (158-166) peptide is a specific peptide-major histocompatibility complex (pMHC) that serves as a critical target for cancer immunotherapy. NY-ESO-1, also known as Cancer-testis antigen 1B (CTAG1B), is a highly immunogenic protein expressed in various malignancies, including synovial sarcoma and melanoma, but its expression in healthy tissues is restricted to the testes (PMID: 9050054). The specific epitope LLMWITQCF (amino acids 158-166) is processed and presented by the HLA-A*24:02 allele, which is highly prevalent in East Asian populations (PMID: 15150592). This pMHC complex is recognized by the T-cell receptors (TCRs) of CD8+ cytotoxic T lymphocytes, initiating a targeted immune response against the tumor. Therapeutic strategies targeting this complex primarily involve TCR-engineered T-cell (TCR-T) therapies, such as TBI-1301, which are designed to bind the pMHC with high affinity and specificity (PMID: 30104344). These treatments aim to exploit the restricted expression of NY-ESO-1 to eliminate cancer cells while minimizing damage to normal tissues. However, challenges such as potential cross-reactivity with similar self-peptides and the risk of cytokine release syndrome remain significant considerations in clinical development (PMID: 25199795).
