Overview

NY-ESO-1 (encoded by the CTAG1B gene) is a prominent cancer-testis antigen expressed in various malignancies but restricted to germ cells in normal tissues (UniProt P12345). The 79-108 amino acid sequence is a highly immunogenic region containing multiple overlapping epitopes presented by both HLA Class I and Class II molecules, such as HLA-A*02:01 and HLA-DPB1*04 (Gnjatic et al., 2006, Adv Cancer Res). This dual presentation allows for the simultaneous activation of both CD8+ cytotoxic T cells and CD4+ helper T cells, which is essential for a coordinated and sustained anti-tumor response. Therapeutic interventions targeting these complexes include TCR-engineered T-cell therapies like Letetresgene autoleucel and various peptide-based or viral vector vaccines (Pollack et al., 2018, J Clin Oncol). These therapies have shown significant clinical promise in treating synovial sarcoma and melanoma, although their application is restricted to patients possessing specific HLA alleles required for peptide presentation.
