Overview

The Human Leukocyte Antigen DP4 (HLA-DP4) presenting NY-ESO-1 (157-170) peptide complex is a specific peptide-major histocompatibility complex (pMHC) class II target. HLA-DP4, specifically the DPA1*01:03/DPB1*04:01 heterodimer, is one of the most prevalent MHC class II alleles, found in approximately 70% of Caucasians (PMID: 11160677). The NY-ESO-1 protein (encoded by CTAG1B) is a well-characterized cancer-testis antigen that is highly expressed in various malignancies but absent in normal adult tissues except for the testis and placenta (PMID: 9031856). The 157-170 epitope (SLLMWITQCFLPVF) is a dominant CD4+ T-cell epitope that binds with high affinity to the HLA-DP4 groove (PMID: 10946282). Recognition of this pMHC complex by specific T-cell receptors (TCRs) triggers CD4+ T-cell activation, which provides essential help for CD8+ T-cell responses and can exert direct anti-tumor effector functions (PMID: 16339512). This complex is a primary target for TCR-engineered T-cell therapies and cancer vaccines aimed at treating NY-ESO-1-positive tumors such as synovial sarcoma and melanoma (PMID: 25731168).
