Human leukocyte antigen class II (HLA-II) molecules are cell-surface glycoproteins encoded by the major histocompatibility complex (MHC) on chromosome 6. They play a central role in the adaptive immune system by presenting processed extracellular antigens to CD4+ T helper cells, thereby initiating and regulating immune responses. They are heterodimers composed of an α and β chain, with the peptide-binding groove formed by the α1 and β1 domains. The main classical HLA class II proteins are HLA-DP, HLA-DQ, and HLA-DR. HLA-II polymorphisms are associated with susceptibility or resistance to autoimmune diseases and transplant compatibility.
