Overview

Human lncRNA metabolic regulator 1 (hLMR1) is a hepatocyte-enriched long non-coding RNA involved in the transcriptional regulation of genes controlling hepatic lipid and cholesterol synthesis. It operates primarily by recruiting the RNA-binding protein PTBP1 to the promoters of cholesterol biosynthesis genes, thus enhancing their transcription. hLMR1 also binds to PABPC1, suppressing protein translation, which in turn impacts the expression of lipogenic enzymes and overall lipid metabolism. Elevated levels of hLMR1 are linked to the development of nonalcoholic fatty liver disease and hyperlipidemia, and specific genetic variants in its locus (such as the SNP rs9653945 major allele 'G') further increase susceptibility to these conditions. Current research highlights its central role in disease-associated metabolic reprogramming of the liver but indicates that hLMR1 is not yet a direct therapeutic target; further studies are needed to develop pharmacological modulators or biomarkers for clinical use.
