Overview

Human neutrophil antigens (HNA) represent a diverse group of glycoproteins expressed on the surface of neutrophils that are polymorphic within the human population. These antigens are categorized into five distinct systems (HNA-1 through HNA-5) based on the specific proteins they reside upon, such as Fc gamma receptor IIIb (CD16b), CD177, and various integrins. While they serve physiological roles in immune cell adhesion and signaling, their primary clinical significance lies in their role as targets for alloantibodies. These antibodies can trigger serious adverse reactions, including transfusion-related acute lung injury (TRALI) and neonatal alloimmune neutropenia (NAN). Because the term refers to a collective group of distinct proteins rather than a single molecular entity, it is generally classified as a category of biomarkers or clinical triggers rather than a specific therapeutic drug target.
