Human papillomavirus E5 protein is a small, highly hydrophobic, membrane-bound viral protein, ranging in size from 40 to 85 amino acids depending on genotype (e.g., HPV-16 E5 is 83 amino acids). E5 is expressed during early phases of the HPV life cycle and is associated primarily with membranes of the endoplasmic reticulum, Golgi apparatus, and nuclear envelope. While its expression in cancers is often lower than that of E6 or E7, E5 contributes to oncogenesis by: - Activating the epidermal growth factor receptor (EGFR) via interfering with its downregulation, stimulating cell proliferation; - Disrupting cell cycle control, including inhibition of tumor suppressor p21; - Promoting immune evasion and escape from host immunological surveillance; - Inducing ER stress and apoptosis regulation. E5's structure is mainly defined by transmembrane α-helices, and it lacks enzymatic activity, functioning through interactions and modulation of cellular proteins. It is conserved in high-risk HPV genotypes and represents a potential antigen and therapeutic target, especially for vaccines. E5 should be considered an HPV oncoprotein and is relevant to cervical and other HPV-associated cancers. Extensive research continues to clarify its mechanisms and potential utility in therapeutic intervention.
