Overview

The human papillomavirus E6 oncoprotein is an approximately 150–160 amino acid protein encoded by the early region of high-risk HPV genomes such as HPV16 and HPV18[1][2][4][7][8]. E6 is a key driver of malignant transformation: it binds the cellular E3 ubiquitin ligase E6AP (UBE3A) to form a complex that targets the tumor suppressor p53 for ubiquitin-mediated degradation, thus impairing apoptosis and cell cycle checkpoints[2][4]. E6 also interacts with PDZ domain-containing proteins (MAGI-1, SAP97/Dlg) at cell junctions, targeting them for proteasomal degradation and promoting loss of cell polarity and further oncogenic activity[3][5][6]. Beyond these classical functions, E6 modulates telomerase activation, gene expression, DNA damage repair, and cellular signaling, enabling persistent infection and tumor development[1][8]. E6, along with E7, is the principal molecular target in the development of new therapeutics for HPV-related cancers thanks to its essential role in cell transformation and malignancy[1].

Mechanism of action

Inhibition of E6-E6AP-p53 complex formation: Some experimental molecules block E6 binding to E6AP, preventing p53 degradation. Disruption of E6-PDZ domain binding: Inhibitors aim to prevent E6-mediated degradation of PDZ domain tumor suppressors. Gene silencing: RNA antagonists target E6 mRNA to reduce its expression.
