Overview

The Human papillomavirus (HPV) E7-derived peptide-MHC complex is a critical immunological target in the treatment of HPV-associated malignancies, such as cervical and oropharyngeal cancers. The E7 protein is a primary oncoprotein that drives malignant transformation by binding and degrading the retinoblastoma protein (pRb), leading to uncontrolled cell cycle progression. In the context of immunotherapy, dendritic cells (DCs) process E7 proteins into short peptides and present them via Major Histocompatibility Complex (MHC) Class I and Class II molecules to prime CD8+ and CD4+ T cells, respectively. This presentation is the foundational step for generating a robust anti-tumor cellular immune response. Therapeutic strategies targeting this complex include synthetic long peptide vaccines, DNA vaccines, and adoptive T-cell therapies using T-cell receptors (TCRs) engineered to recognize specific E7 epitopes (e.g., E7 11-20 or 82-90) presented by HLA-A*02:01. By enhancing the visibility of these viral antigens to the immune system, these treatments aim to selectively eliminate HPV-transformed cells while sparing healthy tissue.
