Overview

The Human Papillomavirus (HPV) Major Capsid Protein L1 is the primary structural component of the HPV virion, responsible for the assembly of the viral shell and facilitating entry into host cells (UniProt P03101). It possesses the unique ability to self-assemble into non-infectious virus-like particles (VLPs) that are morphologically and antigenically identical to the native virus (PubMed, 17012379). These VLPs are the active components in prophylactic vaccines, such as Gardasil 9 and Cervarix, which are designed to stimulate the human immune system (CDC, 2021). Upon administration, the immune system generates high titers of neutralizing antibodies that target the L1 protein, effectively blocking the virus from entering basal epithelial cells during subsequent exposure. This mechanism prevents persistent HPV infection, which is the leading cause of cervical cancer, as well as other anogenital and oropharyngeal malignancies (StatPearls, 2023). By targeting the L1 protein, these vaccines provide long-term protection against the most oncogenic strains of the virus, significantly reducing the global burden of HPV-related diseases.
