Human papillomavirus (HPV) E7 is a critical oncoprotein produced by high-risk HPV types, such as HPV16 and HPV18. As one of only two viral proteins consistently expressed in HPV-associated cancers, E7 plays a central role in both the viral life cycle and carcinogenic transformation. E7 functions primarily as a transcriptional regulator, disrupting the pRb/E2F pathway, targeting numerous cell cycle regulators, interfering with the activity of various cellular transcription factors, interacting with metabolic regulators and associating with histone deacetylases. E7 is essential for the normal virus life cycle and throughout the cancer development pathway. As one of the primary oncoproteins in HPV-associated cancers, E7 is a critical target for diagnostic development for HPV-related malignancies, therapeutic interventions targeting HPV-driven cancers and vaccine development strategies.
