Human Papillomavirus Protein E6 (E6)

Target
E6
Molecular classification
Oncoprotein, Viral protein
01

Overview

The E6 protein is an oncoprotein encoded by high-risk human papillomavirus (HPV) types, notably HPV-16 and HPV-18. It plays a central role in the induction and maintenance of cellular transformation, contributing directly to the development of cervical cancer and other HPV-associated malignancies. E6 is essential for viral replication, persistence of infection, and evasion of host cell defenses. The hallmark function of high-risk HPV E6 is targeting the tumor suppressor p53 for ubiquitin-mediated proteasomal degradation and interacts with other cellular targets.

02

Mechanism of action

Disrupt E6 interaction with E6AP/p53; Inhibit E6 interaction with PDZ domain proteins; Block E6-mediated apoptosis inhibition; Target E6 for degradation
