Overview

Human papillomavirus (HPV) replication and oncoprotein functions encompass the coordinated activities of viral proteins required for the maintenance of the viral episome and the induction of cellular proliferation. The E1 and E2 proteins are primary regulators of viral DNA replication and transcription, while the E6 and E7 oncoproteins are the principal drivers of HPV-induced malignancy (Doorbar et al., 2012). E6 facilitates the ubiquitination and degradation of the tumor suppressor p53, and E7 binds and inactivates the retinoblastoma protein (pRb), leading to uncontrolled cell cycle progression (Hoppe-Seyler et al., 2018). Broad, non-specific interference refers to therapeutic strategies that disrupt these processes collectively, such as through the use of immunomodulators like Imiquimod or broad-spectrum antivirals like Cidofovir (StatPearls, 2023). These interventions aim to halt viral persistence and reverse the oncogenic signaling pathways that lead to cervical, oropharyngeal, and other anogenital cancers (PubMed, PMID: 29331989).
