Overview

Human papillomavirus type 16 and type 18 are the two most clinically important high-risk HPV viruses, together responsible for approximately 70% of all cervical cancers and a significant proportion of other anogenital and oropharyngeal cancers[7]. Their genomes encode several viral proteins including *E6, E7, E5,* and *E1* that manipulate host cell regulatory machinery, disrupt immune signaling, suppress apoptosis, and trigger malignant transformation[1][3][6][7]. The E6 protein induces degradation of p53, while E7 neutralizes pRb, leading to loss of cell cycle control and immortalization. Both are established molecular targets for novel therapies and biomarkers for diagnostic purposes, while current preventive vaccines target the L1 capsid protein to block infection[4][7]. Although "HPV-16 and HPV-18" itself does not refer to a single molecule, their encoded proteins (especially E6, E7) are canonical molecular targets in HPV-associated disease settings.

Mechanism of action

Vaccines: induce neutralizing antibodies against L1 capsid protein to prevent infection. RNA interference, small molecule inhibitors: suppress E6/E7 activity, restore p53/pRb tumor suppressor functions. Immunotherapeutic approaches: enhance immune rejection of HPV-transformed cells.
