Overview

The Human papillomavirus type 16 (HPV16) E6 and E7 oncogenic proteins are the primary drivers of malignancy in high-risk HPV infections. These viral proteins function by systematically dismantling host cell surveillance mechanisms; E6 recruits the E6AP ubiquitin ligase to target the tumor suppressor p53 for proteasomal degradation, while E7 binds to and inactivates the retinoblastoma protein (pRb), thereby releasing E2F transcription factors to drive the cell cycle [1, 3, 13]. Together, they promote uncontrolled cellular proliferation, inhibit apoptosis, and induce genomic instability, leading to the development of cervical, oropharyngeal, and other anogenital cancers [5, 12, 15]. Because E6 and E7 are constitutively expressed in cancerous cells but absent in normal tissues, they represent ideal targets for therapeutic intervention [1, 9]. Current drug development efforts focus on therapeutic vaccines, such as DNA and peptide-based formulations, as well as adoptive T-cell therapies designed to elicit a robust immune response against these oncoproteins [1, 15]. Experimental approaches also include small molecule inhibitors and CRISPR-Cas9 mediated gene silencing to restore host tumor suppressor functions [1, 8, 9]. These therapies aim to selectively eliminate HPV-transformed cells while sparing healthy tissue, addressing a significant unmet need in HPV-associated oncology.
