Overview

The HPV16 E6 oncogene is a primary driver of malignancy in high-risk human papillomavirus (HPV) infections, particularly in cervical and oropharyngeal cancers (1.1.1, 1.5.2). It encodes the E6 oncoprotein, which functions by recruiting the host E3 ubiquitin ligase E6AP to target the tumor suppressor p53 for proteasomal degradation, thereby bypassing cell cycle checkpoints and inhibiting apoptosis (1.3.2, 1.5.1). Additionally, E6 activates telomerase (hTERT) to promote cell immortalization and interacts with various cellular proteins to disrupt epithelial organization and evade the host immune response (1.5.1, 1.5.5). Because the E6 gene is constitutively expressed in HPV-transformed cells but absent in healthy tissues, it is an ideal therapeutic target (1.1.1, 1.2.2). Current therapeutic approaches include DNA vaccines like VGX-3100 and viral-vector vaccines like PRGN-2009, which aim to stimulate a T-cell mediated immune response against infected cells (1.2.1, 1.2.3). Emerging technologies such as CRISPR-Cas9 and TALENs are also being explored to directly edit and disrupt the E6 DNA sequence to halt oncogenic progression (1.2.1, 1.3.3).
