Human papillomavirus (HPV) type 16 and 18 E7 oncoproteins are essential viral proteins responsible for the development and maintenance of high-risk HPV-associated cancers, including cervical, oropharyngeal, and anal carcinomas (Source: NIH, UniProt P03129). These proteins function as oncoproteins by binding to the retinoblastoma tumor suppressor protein (pRb), triggering its ubiquitin-mediated degradation and subsequently disrupting cell cycle checkpoints to promote uncontrolled cellular proliferation (Source: PubMed PMID: 29115210). Because E7 expression is restricted to infected and transformed cells and is absent in healthy human tissue, it represents an ideal tumor-specific antigen for therapeutic targeting. Current clinical strategies focus on the elimination of E7-expressing cells through therapeutic vaccines, such as VGX-3100 and ISA101, and adoptive T-cell therapies (TCR-T) that utilize engineered T-cells to recognize E7 peptides presented on the cell surface (Source: ClinicalTrials.gov). These interventions aim to restore immune surveillance and induce the regression of HPV-induced lesions and tumors.
Drugs targeting HPV16 and HPV18 E7-expressing cells primarily utilize immunotherapy to induce or provide a T-cell mediated attack against the viral oncoproteins. Therapeutic vaccines (e.g., DNA or peptide-based) stimulate the endogenous immune system to recognize E7 epitopes, while adoptive T-cell therapies (TCR-T) involve the infusion of T-cells engineered with high-affinity receptors specific for E7 peptides presented by MHC Class I molecules. Biologically, the E7 protein acts by binding to and degrading the retinoblastoma protein (pRb), which leads to the release of E2F transcription factors and the promotion of the S-phase of the cell cycle, driving oncogenic transformation.
