Overview

Human papillomavirus type 16 (HPV16) L1 major capsid protein conformational epitopes are the primary antigenic determinants recognized by the host immune system to neutralize the virus (NIH, 1996; NIH, 2004). The L1 protein spontaneously self-assembles into pentameric capsomeres, which further organize into icosahedral virus-like particles (VLPs) that mimic the native virion's surface (UniProt, 2016; NIH, 2012). These conformational epitopes, primarily located on the surface-exposed loops (BC, DE, EF, FG, and HI), are essential for the induction of high-titer neutralizing antibodies (NIH, 2025; NIH, 2004). Prophylactic vaccines, such as Gardasil and Cervarix, utilize recombinant L1 VLPs to elicit an immune response against these specific epitopes, effectively preventing HPV16 infection (CDC, 2024; CDC, 2025). Biologically, the L1 protein mediates the initial attachment of the virus to heparan sulfate proteoglycans on the basement membrane or cell surface, a process that triggers conformational changes necessary for viral entry (UniProt, 2016; NIH, 2026). Persistent infection with HPV16 is strongly associated with the development of cervical, anogenital, and oropharyngeal cancers, making these epitopes critical targets for cancer prevention (NIH, 1997; NCI, 2021).
