Overview

The E6 protein is an oncoprotein encoded by the high-risk human papillomavirus type 16 (HPV16). It promotes cellular immortalization and plays a central role in cervical carcinogenesis and other HPV-associated malignancies by targeting key tumor suppressors like p53 and regulators such as hTERT. It contains two zinc finger domains and a C-terminal PDZ-binding motif.

Promotes p53 degradation via ubiquitin-dependent proteasomal pathway and E6AP recruitment; interacts with CBP/p300 to repress transcription; activates telomerase; disrupts cell polarity via PDZ domain interactions; enhances translation initiation complex assembly.
