Overview

The Human papillomavirus type 16 (HPV16) oncoprotein E7 is a multifunctional viral protein that plays a central role in the HPV life cycle and in the development of HPV-associated cancers, particularly cervical cancer. It drives quiescent cells into the cell cycle by stimulating progression from G1 to S phase, facilitating viral genome replication. E7 binds to retinoblastoma tumor suppressor protein (pRB), as well as related pocket proteins p107 and p130, leading to their degradation. It also interacts with cullin 2 ubiquitin ligase complex contributing to pRB degradation and interacts with additional targets such as histone deacetylases and the large retinoblastoma-associated factor p600.
