Human Papillomavirus Type 16 Protein E6 (E6)

Target
E6
Molecular classification
Viral Oncoprotein
01

Overview

Human papillomavirus type 16 (HPV16) protein E6 is a viral oncoprotein encoded by the early region of the HPV16 genome, one of the most prevalent high-risk HPVs causally related to anogenital and head & neck malignancies. E6 acts primarily as an oncoprotein, playing a major role in cellular transformation by binding to host ubiquitin-protein ligase UBE3A/E6AP and targeting tumor suppressors TP53 (p53) and TP73 for proteasomal degradation. It also promotes degradation of other cellular targets and inhibits immune responses.

02

Mechanism of action

Current research focuses on directly inhibiting or silencing E6 oncoproteins as potential therapies against cervical cancer cells.

05

Safety considerations

Targeting E6 can have off-target effects due to its interaction with multiple host cell proteins.Resistance mechanisms may develop with prolonged E6 inhibition.
