The E7 protein is a critical oncoprotein encoded by high-risk human papillomavirus type 18 (HPV18). It plays a central role in the viral life cycle and is essential for the virus's ability to manipulate host cellular processes, promote uncontrolled cell growth, and contribute to cancer development. E7 acts as a master regulator of cellular disruption, primarily by binding and degrading the retinoblastoma protein (pRb), leading to the release and activation of E2F transcription factors and uncontrolled cell cycle progression. It also promotes genomic instability, inhibits apoptosis, and evades the immune system. It's particularly associated with adenocarcinomas compared to other high-risk types like HPV16. E7 is a phosphoprotein, and its phosphorylation status affects its stability. Studying HPV18 E7 provides insights into cancer development and opportunities for therapeutic targeting, especially through vaccines or inhibitors aimed at blocking its interaction with pRb or other host proteins.
