Overview

Human Papillomavirus type 33 (HPV33) major capsid protein L1 is the primary structural component of the viral capsid, which forms an icosahedral shell composed of 72 pentameric capsomeres (UniProt P06416). This protein has the unique ability to self-assemble into virus-like particles (VLPs) that are antigenically and morphologically similar to the native virion but lack the oncogenic viral genome, making them ideal for vaccine development (PubMed: 25410398). Biologically, the L1 protein mediates the initial infection of basal epithelial cells by binding to heparan sulfate proteoglycans on the basement membrane (PubMed: 18037307). HPV33 is recognized as a high-risk genotype significantly associated with the development of cervical, anogenital, and oropharyngeal cancers (WHO/IARC). From a pharmacological perspective, the HPV33 L1 protein is a critical component of the 9-valent HPV vaccine (Gardasil 9), which utilizes the protein's immunogenicity to elicit neutralizing antibodies (FDA Label: Gardasil 9). Prophylactic immunization targeting this protein prevents the establishment of persistent HPV33 infection, thereby reducing the subsequent risk of developing HPV-related malignancies.
