Overview

The Human papillomavirus type 58 E6 protein is an oncogenic viral protein expressed by the high-risk HPV58 genotype, which is associated with cervical cancer and other anogenital malignancies[1][3]. E6 contains two zinc finger domains formed by Cys-X-X-Cys motifs and has critical functions in cellular transformation. Its key oncogenic mechanism involves binding the cellular ubiquitin ligase E6AP (UBE3A) via an LXXLL consensus motif, forming a trimeric complex with the tumor suppressor protein p53. This interaction triggers ubiquitin-mediated degradation of p53, abolishing cell cycle arrest and apoptosis, and permitting unchecked cell proliferation[2][3]. E6 also interacts with a wide range of cellular partners through a C-terminal PDZ-binding motif, disrupting signaling and cell polarity. Beyond p53, E6 impedes other apoptotic pathways, modulates mTOR signaling, and blocks cellular senescence. The protein is the subject of therapeutic drug discovery and biomarker research, but clinically approved E6-specific drugs are not currently available[2][3][1].

Mechanism of action

Inhibitors act by blocking E6’s binding to E6AP or the LXXLL motif, preventing p53 degradation. Restoration of p53 function by disruption of E6-E6AP-p53 complex.
