Overview

The Human papillomavirus type 58 (HPV58) L1 major capsid protein is the primary structural component of the HPV58 virion, an icosahedral virus belonging to the Alphapapillomavirus genus. HPV58 is classified as a high-risk genotype and is notably more prevalent in East Asian populations compared to other regions, where it is a significant driver of cervical cancer and its precursor lesions (UniProt P26538; PubMed: 25135154). The L1 protein has the intrinsic ability to self-assemble into virus-like particles (VLPs), which are morphologically and antigenically similar to the native virus but lack the viral genome (PubMed: 30243934). These VLPs serve as the active immunogen in the 9-valent HPV vaccine, inducing a robust humoral immune response characterized by the production of type-specific neutralizing antibodies (FDA: Gardasil 9 Label). These antibodies bind to the L1 protein on the surface of the virus, preventing its interaction with host cell receptors such as heparan sulfate proteoglycans and thereby blocking viral entry into the basal layer of the epithelium (PubMed: 23935941). While highly effective for prophylaxis, the L1 protein is not typically expressed in late-stage cancerous cells where the viral genome is integrated, making it a target for prevention rather than treatment of established malignancy.
