Overview

The Human papillomavirus type 6 E6 protein (HPV6 E6) is a viral oncoprotein essential for the maintenance and replication of the HPV6 genome in host epithelial cells (UniProt: P06462). Although HPV6 is classified as a low-risk genotype because its E6 protein does not efficiently induce the degradation of the p53 tumor suppressor like high-risk types, it still interacts with various host proteins to delay terminal differentiation and promote cell proliferation (PubMed: 10482588). This protein is a primary driver of diseases such as condyloma acuminatum (genital warts) and recurrent respiratory papillomatosis (RRP), where its continuous expression is required for the persistence of benign tumors (PubMed: 30535119). Because E6 is expressed throughout the viral life cycle, it serves as a critical target for therapeutic vaccines, including DNA-based (INO-3106) and adenoviral-vector (PRGN-2012) platforms (ClinicalTrials.gov: NCT04398420). These immunotherapies are designed to elicit a robust T-cell mediated response to clear persistent infections that are otherwise resistant to traditional surgical interventions. Successful targeting of HPV6 E6 aims to provide a non-invasive solution for chronic HPV6-related pathologies and reduce the high rates of lesion recurrence.
