Overview

The Human papillomavirus type 6 (HPV6) E6 protein-derived peptide-MHC complex is a specialized immunological target consisting of short viral peptide fragments bound to Major Histocompatibility Complex (MHC) molecules on the surface of infected cells. HPV6 is a low-risk HPV genotype primarily responsible for benign but morbid conditions such as genital warts and recurrent respiratory papillomatosis (RRP). The E6 oncoprotein is constitutively expressed in HPV-infected cells and plays a critical role in maintaining the viral life cycle and promoting cellular proliferation, albeit with lower oncogenic potential than high-risk HPV types. Therapeutic strategies targeting these peptide-MHC complexes aim to bypass the lack of natural robust immunity by using DNA vaccines, viral vectors, or TCR-engineered T-cells to prime the immune system. When a T-cell receptor (TCR) recognizes the specific HPV6 E6 peptide presented by the MHC, it triggers a cytotoxic response that leads to the apoptosis of the infected cell. This target is particularly valuable for treating chronic or recurrent HPV6-related lesions where surgical intervention is often insufficient due to high recurrence rates. Current clinical candidates like INO-3106 and PRGN-2012 are designed to elicit these specific T-cell responses to clear persistent viral reservoirs.
