Overview

The Human papillomavirus type 6 L1 protein is a ~55 kDa viral capsid protein that forms pentameric capsomers, which self-assemble to create the exterior shell of HPV6 virions. Each virion is composed of 72 pentamers of L1, arranged in an icosahedral structure stabilizing both the viral genome and capsid integrity. The L1 surface loops display the highest sequence variability, serve as neutralizing epitopes, and are the main target of immune responses induced by HPV vaccines. L1 mediates initial viral binding to host cell surface receptors (primarily heparan sulfate proteoglycans and laminin-332), initiating infection. Recombinant expression of L1 yields virus-like particles (VLPs) that are non-infectious but immunogenic, forming the basis for prophylactic HPV vaccines. HPV6 is classified as a "low-risk" type associated with benign lesions such as genital warts and respiratory papillomatosis, without oncogenic potential.
