Overview

The Human papillomavirus type 6 (HPV6) E2 protein is a key regulatory factor essential for the viral life cycle of low-risk HPV types. It functions as a sequence-specific DNA-binding protein that regulates the transcription of viral genes, most notably the E6 and E7 oncogenes, and is indispensable for viral DNA replication (UniProt P03120). By forming a complex with the E1 helicase, E2 facilitates the recruitment of the replication machinery to the viral origin of replication (PubMed: 15134133). HPV6 is the primary causative agent of genital warts and recurrent respiratory papillomatosis, making its proteins significant therapeutic targets (NIH: StatPearls). Therapeutic strategies focusing on E2 involve the development of small molecules that disrupt the E1-E2 protein-protein interaction or inhibit E2's ability to bind to viral DNA. While no E2-specific inhibitors are currently FDA-approved, research into indandione and benzothiazepine derivatives has shown promise in preclinical models for reducing viral load and lesion growth.
