Human pluripotency-associated transcript 5 (HPAT5) is a primate-specific long noncoding RNA that is highly expressed in human embryonic stem cells and is required for the maintenance of pluripotency and proper development of preimplantation embryos. HPAT5 acts through post-transcriptional mechanisms, most notably by binding and sequestering mature let-7 microRNAs—this interaction attenuates let-7 activity and thereby helps preserve stem cell identity and delay differentiation. Loss of HPAT5 leads to increased let-7 levels, which accelerates stem cell differentiation. HPAT5 can function as a biomarker for pluripotency but is not a conventional protein target for drugs[2][3].
