The Human T-cell receptor (TCR) recognizing Respiratory Syncytial Virus (RSV) fusion (F) protein-derived peptides is a specialized immune receptor that mediates the recognition and elimination of RSV-infected cells (PubMed: 31413125). These TCRs specifically bind to viral peptides, such as the immunodominant F85-93 epitope, when presented by Major Histocompatibility Complex (MHC) class I molecules, most commonly HLA-A*02:01 (PubMed: 11160692). Upon engagement with the peptide-MHC complex, the TCR triggers a signaling cascade through the CD3 complex, leading to the activation of cytotoxic T lymphocytes (CTLs), secretion of pro-inflammatory cytokines like IFN-gamma, and direct lysis of target cells. In therapeutic contexts, these TCR sequences are being utilized to engineer TCR-T cells for adoptive immunotherapy, particularly for immunocompromised individuals who cannot mount an effective endogenous response (PubMed: 33854510). The development of these therapies requires precise characterization of TCR affinity and specificity to prevent off-target effects against self-peptides. Furthermore, these receptors are central to the evaluation of T-cell-mediated vaccine efficacy, serving as a benchmark for protective cellular immunity against RSV (PubMed: 28250128).
The T-cell receptor specifically binds to RSV F-derived peptides presented by MHC molecules on the surface of infected cells, triggering T-cell activation, cytokine production, and the targeted destruction of the virus-infected cell (PubMed: 31413125).
