Human telomerase reverse transcriptase (hTERT) promoter-driven selective viral replication is a strategy in which the promoter region of the hTERT gene is used to control the expression of key viral genes, enabling viruses—such as oncolytic adenoviruses or herpes simplex viruses—to replicate preferentially in cancer cells rather than normal cells. This approach exploits the fact that hTERT, and thus telomerase activity, is highly upregulated in most human cancers but remains repressed in normal somatic tissues.
Selectively replicates viruses in cells with high hTERT promoter activity, leading to cell lysis and tumor destruction.
